Method development in liquid chromatography (LC) encompasses the search for the optimal operating conditions (type of mobile and stationary phase, temperature, gradient steepness, pH, ionic strength) resulting in the separation of a mixture into its individual constituents.
Because of the high probability of peak overlap in complex mixtures and the dependence of retention and selectivity on the chromatographic parameters used, the method development process is often tedious and time-consuming (up to several weeks of work).
