We provide a highly efficient high-throughput strain improvement program, which can significantly increase the yield of your target product, or improve the impurity profile of produced target compounds.
ACIES BIO uses two approaches to strain improvement:
- By mutagenesis and high-throughput strain selection using a combination of classical and rational mutagenesis as well as protoplast fusion approaches
- By genetic/metabolic engineering of target pathways, using over-expression of "positive" genes and clusters and/or inactivation of competing or undesired metabolic pathways.
Our strain improvement projects are generally supported by whole-genome analysis and bioinformatics support, to identify themetabolic potential and existing bottlenecks, which can be essential for efficient yield increase. Whole-genome analysis also provides additional safety in strain development as well as in IP support.
Classical Approach – strain selection
- Classical mutagenesis and high-throughput strain selection and analytics
- Directed mutagenesis by metabolic pressure
- Scale-down process optimization
- Starting from existing technologies or from wild type strains
Genetic Approach – metabolic engineering
- Increasing precursor supply
- Up / down-regulation of genes and/or pathways
- Removing competing metabolic pathways and impurities
